All the current Modes and what we know

Asynchronous Online Multiplayer
"Borrow" a friend's character stats for a certain amount of time (your friend will get a bonus). Ghost Data (See friend's saved character movements while fighting a boss). Friend's playthroughs affect your game in some way (e.g. a gravestone and an offering might mark the spot where your friend died). Plus another surprise that we'll unveil soon...Of course we're still in the planning phase, so who knows what we'll end up with.
Online Challenge Mode
Compete and cooperate with a friend to earn points in specially designed stages by clearing challenges, killing enemies, collecting items, and more. Rumor has it there there may even be incredibly powerful bosses which drop rare crafting materials.
New Game +
Next game (of choice) Carry over key items/equipment to your next game. Harder enemies, better stats, and new attack patterns. The team hopes to implement some special enemies which may drop rare items.
Speed Run Mode
No information on this yet, we believe it is about having no cutscenes and a visible timer on screen from start to finish. Features to help speedrunners plan the perfect route.
Classic Mode
This is a cool one, especially if you've been playing action platformers since controllers had two buttons: In Classic Mode, existing assets and screens from the game will be rearranged linearly, to reward timing and survival instead of backtracking and exploration. Six punishing stages. In grand classic platformer fashion, you'll be weaker and the enemies will be stronger, and forget about saving yourself by grinding—no RPG elements here. Old School bit-music.
Prequel Mini-Game
A 2D, pixel-based, retro-styled mini-game that sheds some light on the world of Bloodstained before Miriam awoke from her coma. This is a complete game which will be sold separately, but every $28+ backer copy of Bloodstained will come with a FREE download of this game. (Backers at Physical+Digital tiers will be eligible for 2 copies of the mini-game.) Just a theory: We play as Zangetsu, slaying demons just days before Miriam wakes up.
Retro Level
One section of the castle will feature an area that Miriam can traverse in beautiful retro graphics. We've been referring to this section as the “8-bit Level” internally, although it may be closer to 16+ bit depending on how we mix it with the full game. While a full-blown 8-bit game doesn't fit into the game's budget, backer support will allow us to have fun with the development and create a little retro magic in our game.
Rougelike Dungeons
Procedural dungeons, sharable seeds. Should not be confused with procedural generated backdrops that also are in the game.
Boss Rush Mode
No information on this mode yet. We believe it is room after room, with a timer for you to kill each and every boss. Theories of endless bosses that grows stronger during each playthrough. No mention about level, gear or pause function. IGA Boss not included, (unless if you have not gotten the $60+ dollar tier(s).. perhaps.)
Boss Revenge Mode
Playable bosses versus the heroes in a new area.
Nightmare Difficulty (Mode)
No information provided, we can expect a harder difficulty in one or many different ways. Theory: Rearranged position of enemies. More aggressive enemies. Rearranged items on map (perhaps even with a minor random generated effect of placement). Some items or skills might be locked for our hero(es) to use.
Note: All three playable characters might take part in all the modes, but there is no confirmation. Miriam will not be playable in the Prequel Mini-Game but a theory is that we will play Zangetsu during his demonic hunting, days before Miriam wakes up.
