I decided to calculate the total play area of each Metroidvania I've played. Which is most, if not all of them. They were counted by getting a map of each game, placing a dot in the exact center of each room (a lot of cut/paste was involved to make this easier on my sanity), and using the Magic Wand tool in paint.net to select all of the dots. This showed me how many pixels I had in total. I have come up with these results, sorted by size.
Symphony of the Night: 1,890 squares
Portrait of Ruin: 1,846 squares
Harmony of Dissonance: 1348 squares
Order of Ecclesia: 1,335 squares
Dawn of Sorrow: 1023 squares
Circle of the Moon: 980 squares
Aria of Sorrow: 928 squares
Any room that did not count towards the total % was not included. This shows that Symphony still has the largest total play area, but when you ignore inverted shenanigans, it is beaten out by every other Metroidvania released after it with the exception of Aria of Sorrow which is only a handful of rooms behind.

An intrepid poster in 2014 actually went ahead and calculated the map size of the various Metroidvania games. That means, we can quantify what i means for Bloodstained to be "Iga's Biggest Castle Yet".
