Hm, if I could pick any Castlevaina boss to play as, it would be Walter Bernhard. You could design a whole game around him, technically. His origin, how he came across the Ebony stone, gathering of a legion of monsters, fighting crusaders and other beasts for territory, and set up shop in the Eternal Night Forest. Final boss can be whatever, a former head vampire, The Forgotten One, or even Death. Then, the after credit scene could be set up many years later to when Walter Met Mathias and set up events for Lament of Innocence. Ah, if only, if only...

I've always wanted to play as Death, maybe the one from Dawn of Sorrow. He just looks cool. Also, I wanted to play as Dracula in Order of Ecclesia. If I am able to use Demonic Megiddo, I will abuse it.

Another vote for Dracula. The Dracula X/Symphony of the Night/Nocturn in the Moonlight incarnation.
A little bit of gradual humanity, power, knowledge he cannot die due to third party interference... Would make for an interesting plot.
Death would be another. Especially his and Dracula's comradery for the plot.
And Shaft. An actual full blown mage would be an interesting change of pace.
